Item Creation of anti-inflammatory pharmaceutical compositions (review)(2022-11-13) Syrova, Ganna; Petyunina, Valentina; Makarov, Volodimir; Lukіаnova, Larysa; Chalenko, NataliaThe review article summarizes the data of literature and our own research on the creation of more effective and safe domestic combined drugs with a pronounced anti-inflammatory and analgesic effect. Untimely treatment of inflammatory processes often leads to long-term incapacity and even disability, so today there is an active search for new effective and safe domestic combination drugs with pronounced anti-inflammatory and analgesic effects. Due to the constant search for innovative drugs the treatment of a significant number of diseases and pathological conditions with existing drugs remains ineffective or causes addiction and a large number of side effects. As a result, inflammatory processes can be accompanied by severe pain, spasms, increased convulsive activity of the brain, other disorders, so non-narcotic analgesics and nonsteroidal anti-inflammatory drugs are combined with other agents that can cause complementary effects or potentiate each other's effects. Caffeine is a useful adjuvant of nonsteroidal anti-inflammatory drugs of various chemical structures in terms of anti-inflammatory and analgesic effects. The article presents a literature review of the current state of treatment of inflammation and pain, conducting research to expand the therapeutic capabilities of known pharmaceuticals, the creation of pharmaceutical compositions of nonsteroidal anti-inflammatory drugs with caffeine.Item Experimental investigation of the effect of pharmaceutical composition on the central nervous system(KhNMU, 2019) Syrova, Ganna; Lukianova, Larysa; Sinelnik, Vyacheslav; Krasnikova, Yuliya; Salam, LoginaIt is known that combination of non-steroidal anti-inflammatory drugs with other drugs can cause complementary effects, or potentiate the effects of each other. Object and methods. The objective of this experiment is to analyze the use of a new pharmaceutical composition consisting of nonsteroidal anti-inflammatory drug of the coxib group (rofecoxib) and an immunomodulator of lycopid (glucosaminylmuramyl dipeptide). An experiment on laboratory animals was conducted in order to view the effect of rofecoxib, licopid and their pharmacological composition on the central nervous system and on emotional-behavioral reactions in an "open field" test under formalin edema conditions. Results Previously, we studied the pharmaceutical compositions of nonsteroidal anti-inflammatory drugs of different chemical structure with the psychostimulant caffeine. The results indicated that caffeine potentiated the pharmacological effects of nonsteroidal anti-inflammatory drugs. In this study, we set a goal to create a pharmaceutical composition of rofecoxib with licopid and investigated its effect on the emotional-behavioral reactions of rats under formalin edema conditions. Ànalysis of the experimental results show that the addition of licopid to rofecoxib contributed to an increase in rat's locomotor and orienting-research activities and also an increase in the indicators of their emotional reactions (urination and defecation) compared to the mono-administration of rofecoxib. Conclusions. The pharmaceutical composition of rofecoxib and licopid is expedient and promising for the study of anti-inflammatory and analgesic effects.Item Experimental investigation of the influence of pharmaceutical composition on emotional behavioral reactions under conditions of formalin edema(Cognum Publishing House, Liverpool, United Kingdom, 2019-10-09) Syrova, Ganna; Lukianova, Larysa; Krasnikova, Yuliya; Sinelnik, VyacheslavThe analysis of the results of experimental studies of the effect of celecoxib, licopid and their pharmaceutical composition on the central nervous system by emotional-behavioral reactions in the test "open field" in the conditions of formalin edema indicates that the addition of licopid to celecoxib increased rat’s locomotor and orienting-research activities, the indicators of their emotional reactions (washings and urinations) in comparison with the mono-administration of celecoxib. The pharmaceutical composition of celecoxib and licopid is appropriate and promising for the study of anti-inflammatory and analgesic effects.Item Biochemical Confirmation of Anti-Inflammatory Activity of Oxicam-Based Pharmaceutical Compositions(2018-12-26) Syrova, Ganna; Tishakova, Tetyana; Levashova, Olga; Savelieva, OlenaBiochemical confirmation of anti-inflammatory activity of oxicam-based pharmaceutical compositions was performed by the determination of the level of one of the main markers of inflammation-C-reactive protein. Biochemical studies were carried out on laboratory animals (white WAG rats) to study the anti-inflammatory effects of meloxicam, piroxicam, caffeine, and pharmaceutical compositions consisting of meloxicam and caffeine, piroxicam and caffeine compared to the reference drug - sodium diclofenac. The content of CRP in serum of rats was determined using the CRP latex test kit. It was shown that the composition of meloxicam and caffeine reduced the content of CRP by 16 times compared with formalin-induced edema, and by 2 times in comparison with the reference drug diclofenac sodium, which is statistically significantly different from the control.Item Experimental study of the antipyretic effect of oxicams and their compositions with caffeine(2018-09-30) Syrova, Ganna; Lukianova, Larysa; Kalinenko, Olga; Vodolazhenko, Maria; Petiunina, ValentinaThe experimental study of the influence of caffeine on the antipyretic activity of oxicams in rats was carried out. Meloxicam, piroxicam and their adjuvant caffeine were selected as drugs. According to the results of experimental studies, it was found that meloxicam has the highest antipyretic activity (89,24 %) in 3 hours after administration and exceeded the antipyretic activity of the reference drug. Addition of caffeine to meloxicam did not promote increasing of the meloxicam antipyretic activity, so this composition is considered inappropriate in relation to its antipyretic effect. The antipyretic activity of piroxicam was lower comparing to the reference drug diclofenac sodium. Addition of caffeine to piroxicam promotes increasing of the piroxicam antipyretic activity in 3 hours after administration of the drug. This exceeded the piroxicam antipyretic activity in 1,3 times but did not reach diclofenac sodium activity. The results of experimental studies can become the basis for the creation of new domestic combined medicines.Item The experimental research on the antiinflammatory action of the new piroxicamcaffeine pharmaceutical composition(2018) Syrova, Ganna; Lukianova, Larysa; Chalenko, NataliaConducting an experimental research on laboratory rats subjected to endogastric injections, our scientists have explored the influence of the non-steroid anti-inflamatory drug from the oxicam group piroxicam (4-hydroxo-2-methyl-N-pyridine-2-il)-2H-1,2-benzothiazine-3-carboxamide1,1- dioxyde), a well-known adjuvant of non-narcotic analgetics and non-steroid anti-inflamatory drugs such as caffeine (1,3,7-trimethylxanthine)and the two-component piroxicam-caffeine pharmaceutical composition on the rats’ anti-exudative activity under formalin edema. The analysis of the results of the experimental researches points to the fact that caffeine promotes the anti-exudative action of piroxicam in the rats under formalin edema. The above-mentioned experimental researches allow us to come to the conclusion that due to its vasoconstrictive effect caffeine performs as an adjuvant to piroxicam (a non-steroid anti-inflamatory drug from the oxicam group) concerning the anti-exudative activity. The results of the research can become a basis for the development of new combined drugs in Ukraine.Item The experimental study of pharmaceutical composition piroxicam and caffeine(Scientific Educational Center Warsaw, Poland, 2017-11-18) Syrovaya, Anna; Lukianova, Larysa; Kolesnyk, Maria; Adamu, IsaakaIn an experiment on rats, the effects of a non-steroidal anti-inflammatory agent of the group of oxicams piroxicam, caffeine and their composition on emotional-behavioral reactions under formalin edema were conducted. Evaluation of drug’s and their compositions influence on characteristics of animal’s behavior was done by comparison remedies with control and with formalin edema. Observation of the parameters of routine research activity in the "open field" test and the multi-parameter method for evaluating anxiety-phobic states was carried out according to the generally accepted method. The analysis of the results of experimental studies indicates that caffeine increases the activity of piroxicam relatively to horizontal and vertical motion activity, cognitive activity, grooming and the number of urinations in rats under formalin edema. The results of research can be the basis for the development of new domestic combined medicines.
