Effects of 5 SNPs on daughters' milk performance traits produced by Ukrainian dairy sires
    (2018) Fedota, O.; Lysenko, N.; Mitiohlo, L.; Ruban, S.
The aim of the study was to estimate the effects of the CAPN316, CAST282, GH L127V, GHR F279Y and A257G on milk yield, fat and protein yield and content on daughters produced by dairy sires. 24 dairy sires were 100% Holstein (14), 100% Brown Swiss (5) and Holstein mixed with another dairy breeds (5). Analyses included data on 32733 daughters for 2014-2017 years. Molecular genetic analysis was performed by PCR-RFLP. Statistical analysis revealed that population was on Hardy-Weinberg equilibrium. SNPs F279Y and A257G were not in linkage disequilibrium (r2 = 0.029, D’ = 0.074), because linkage of certain alleles was observed only in 7.4% of cases (D’). C-allele of SNP CAPN316 was strongly associated with protein content, despite C-allele of SNP CAST282 showed negative association with milk, fat and protein yield. Daughter milk performance traits (DMPTs) for SNP CAPN316 corresponded to CC>CG>GG pattern, except variations in milk and fat yield (kg). For SNP CAST282 Milk, fat and protein yield (kg) were significantly higher for CG than for CC genotype. C-allele for SNP L127V showed significant association and differences between genotypes with DYDs for fat yield (kg) – 20.45±5.53 (P<0.01) and content (%) – 0.13±0.06 (P<0.05), T-allele for SNP F279V was not significantly associated with any trait studied and A-allele for SNP A257G significantly associated with milk and fat yield (kg) for DMPTs/DYDs 1988.33±419.93/-269.69±89.92 (P<0.01) and 84.30±15.64/-8.33±3.87 (P<0.05). Results of this study reveal and support known advantages of SNPs studied in panels and selection programs.

The Effects of Polymorphisms in Growth Hormone and Growth Hormone Receptor Genes on Production and Reproduction Traits in Aberdeen-Angus Cattle (Bos taurus L., 1758)
    (2017) Fedota, O.; Lysenko, N.; Ruban, S.; Kolisnyk, O.; Goraichuk, I.
The study was aimed to analyze the relation between individual genotypes and allelic variants of SNPs g.2141C>G of growth hormone gene, g.914T>A and g.257A>G of growth hormone receptor gene with growth and reproduction traits and to evaluate the populationgenetic structure in Aberdeen-Angus cattle (Bos taurus L., 1758) sample of Eastern Ukraine according SNPs studied. Allele C of SNP g.2141C>G has a positive correlation with birth weight, body stature, bigger rump, udder and total exterior evaluation score, shorter calving interval and better calve birth weight and negative correlation with calve average daily gain. Allele T of SNP g.914T>A has positive correlation with the muscle and udder size; live weight in each age, average daily gain, weight and average daily gain of calves born conform to the principle AA>TT≥TA. SNP g.257A>G showed a positive correlation for G allele with muscle size. The population is in equilibrium for SNPs g.2141C>G and g.257A>G, and in disequilibrium for SNP g.914T>A. The analysis showed no linkage disequilibrium between SNPs g.914T>A and g.257A>G. Inbreeding coefficient FST in Aberdeen-Angus group studied was 16.1%.

The effects of polymorphisms in calpain, calpastatin and growth hormone genes on growth traits in Angus cows
    (2016) Ruban, S.; Fedota, O.; Lysenko, N.; Kolisnyk, A.; Goraichuk, I.
The study was aimed to determine the allele and genotype frequencies of SNPs for gene calpain (CAPN316), calpastatin (CAST282) and growth hormone (GH L127V) in Aberdeen-Angus (n=52) and to evaluate their impact on body weight dynamics until age of 5 years. The allele and genotype frequencies was CAPN316: C = 0.45, G = 0.55; CC = 19.2%, CG = 51.9%, GG = 28.9%, CAST282: C = 0.75, G = 0.25; CC = 53.8%, CG = 42.3%, GG = 3.9%, GH L127V: C = 0.34, G = 0.66; CC = 9.6%, CG = 48.1%, GG = 42.3%. There is correlation between the number of C-alleles for GH L127V and increase in body weight from birth till two years. The CAPN316 and CAST282 alleles associated with meat tenderness effect on live body weight increase after two year age. A significant effect of genotype CC CAPN316 is observed at the age of three and four years. It was concluded, that selection aimed to improve meat quality does not lead to a significant reduction in live body weight.
